Course objectives
The main purpose of the course is to provide students with a general overview of the basic techniques of electron microscopy and of the possibilities of processing biological material for electron microscopes.
Learning outcomes
At the end of this course, students should be able to explain the principles and application of the electron microscopy, the history of the development of electron microscopes; to describe the construction of transmission electron microscope (TEM) and scanning electron microscope (SEM); to understand and explain the imaging methods in TEM and SEM; to learn basic techniques of specimen preparation for electron microscopy; to apply the knowledge to the selected samples; to interpret biological structures in the electron micrographs; to have knowledge of artefacts; to acquire the principles of the routine operation of the electron microscope.

  • The theoretical part:
  • an introduction to electron microscopy
  • transmission electron microscope
  • preparative methods for TEM
  • scanning electron microscope
  • preparative methods for SEM
  • electron microscopic cytology
  • The practical part:
  • preparing glass knives
  • preparing grids for ultrathin sections
  • cutting and staining of semi-thin sections
  • preparing samples for SEM
  • individual routine operation of the electron microscope

Colloquium consists of oral exam, evaluation of protocols, determination and assessment of 5 electron micrographs
