- Course objectives
- The aim of the course is to famaliarize students with basic methods of mycorrhizal symbioses research.
- Learning outcomes
- At the end of this practical course, students are able to:
distinguish mycorrhizal symbioses on the base of their morphology and anatomy;
evaluate the effects of mycorrhizal symbiosis on plant growth;
quantify the extent of root colonization and external mycelia growth;
cultivate and propagate the mycorrhizal fungi. - Syllabus
- Mycorrhizal colonization of plant roots: commonly used methods, assessment of root colonization (chemical and microscopic methods), observation of typical mycorrhizal structures.
- Isolation of spores of arbuscular and orchid fungi: isolation techniques, staining by Melzer's reagent, sample preparation, spore description and fungal species determination.
- Mycorrhizal extraradical mycelium: extraction techniques (membrane filtration and inserted membrane technique), vital staining techniques, assessment of extraradical hyphal length (grid-line intersect method and image analysis).
- Long-term cultivation experiment: the aim of the experiment vary with every course run.
