Course objectives
The aim of the course is to famaliarize students with basic methods of mycorrhizal symbioses research.
Learning outcomes
At the end of this practical course, students are able to:
distinguish mycorrhizal symbioses on the base of their morphology and anatomy;
evaluate the effects of mycorrhizal symbiosis on plant growth;
quantify the extent of root colonization and external mycelia growth;
cultivate and propagate the mycorrhizal fungi.
Syllabus
  • Mycorrhizal colonization of plant roots: commonly used methods, assessment of root colonization (chemical and microscopic methods), observation of typical mycorrhizal structures.

  • Isolation of spores of arbuscular and orchid fungi: isolation techniques, staining by Melzer's reagent, sample preparation, spore description and fungal species determination.

  • Mycorrhizal extraradical mycelium: extraction techniques (membrane filtration and inserted membrane technique), vital staining techniques, assessment of extraradical hyphal length (grid-line intersect method and image analysis).

  • Long-term cultivation experiment: the aim of the experiment vary with every course run.
