- Course objectives
- Cytogenetics is one of the oldest branches of genetics. The course provides undergraduate students the general introduction to cytogenetics. Main objectives can be summarized as follows: to understand main aspect of chromosome biology and to learn modern cytogenetic techniques and methods.
- Learning outcomes
- At the end of this course, students will be able to prepare chromosomal samples using standard chromosome banding methods used in conventional cytogenetics. They will also be able to interpret human karyotype. They will understand the principles of FISH technique and be able to prepare samples for it, as well as to analyse the results obtained by digital image analysis. They will be able to explain the basic principles of other molecular cytogenetics methods (mFISH, SKY, CGH, arrayCGH, QF-PCR) and their applications in clinical genetics and oncology.
- Syllabus
- 1.Basis of microscopy. Light microscopy, cytogenetic examples from plant, animal and human chromosomes. 2. Human cytogenetics. Basic techniques of human cytogenetic preparation preparing. Chromosome banding methods: G, C, R, N-banding. Chromosome identification, karyotype, idiogram. The most common human chromosomal aberrations. An International System for Human Cytogenetic Nomenclature (ISCN). 3. Molecular cytogenetics. Fluorescence in situ hybridization (FISH) and its application in prenatal, postnatal and cancer genetics. Basic techniques in FISH preparation making. Using of FISH on buccal swab examples. Application of computer image analysis in human cytogenetics. Preparation od PCR reaction for prenatal QF-PCR technique. 4. Molecular cytogenetics. QF-PCR technique and its application in prenatal and postnatal genetics. Preparation of amplified DNA samples for genetic analyzer. Sample evaluation - samples from patients with known 13, 18, 21, X and Y aneuploidies. Array-CGH technique - teoretical principle and its application in prenatal, postnatal and cancer genetics. Working with software for array-CGH interpretation.
