- Course objectives
- At the end of the course students should be able to:
describe basic principles of plant cell, tissue and organ culture in vitro;
explain the history and development of this cultures until today;
analyze the impact of different types of culture method on the plant genotype preservation and/or change;
propose in vitro methods suitable for plant multiplication, breeding and production. - Learning outcomes

- 1. Historical Background, Conditions of in vitro Culture, Laboratory Design.
- 2. Media Composition and Preparation. Plant Hormones and Growth Regulators.
- 3. In vitro Vegetative Propagation: Micropropagation by Proliferation of Axillary Buds, Adventitious Shoot Proliferation; Initiation of Primary Culture, Subculture.
- 4. Callus Culture: Callus Initiation and Maintenance, Subculture, Organogenesis from Callus.
- 5. Meristem Culture: Apical and Axillary Meristem Isolation and Culture.
- 6. Test-tube Pollination/ Fertilization in vitro: Sterile Pollination of Isolated Placentae with Ovules. Fertiliation of Isolated Ovules and/or Isolated Gametes.
- 7. Embryo Culture: Isolation and Culture of Young and Mature Zygotic Embryos.
- 8. Somatic Embryogenesis: Direct and Indirect Somatic Embryogenesis.
- 9. Ploidy reduction: Androgenesis: Initiation of Anther or Microspore Culture,Chromosome Doubling.
