- Course objectives
- The main aims in the course are a)to acquaint students with the basic techniques of light microscopy and the possibilities of processing biological material in the form of different types of prepared microscope slides; b) to learn students with basic principles of design and analysis of biological experiments; c) give students a short overview of computer-aided image analysis.
- Learning outcomes
- At the end of this practical course, students should be able - to use various techniques of light microscopy efficiently; - to prepare various kinds of botanical or zoological mounts; - to take and efectivelly process microphotographs; - to utilize some simple techniques of image analysis; - to conduct simple biological experiment including adequate statistical treatment of the data; - to utilize basic facts about mitosis, embryogenesis and the other biological processes in their own teaching practice.
- Syllabus
- Introduction. Light microscope: structure and function. Köhler illumination. Botanical techniques: Microrelief technique for the study of the stomata structure and density. Whole mounts of cleared preparations: development of Angiosperm (dicotyledonous plant) embryo. Squash preparations: cell cycle, mitosis, chromosome counting. Anatomical methods: sectioning (hand sections, fixation, infiltration and embedding, microtome Sections), staining. Documentation - hand drawing, microphotographs and their processing. Simple technique of image analysis - leaf area measurement. Design and statistical treatment of simple biological experiments. Zoological techniques: Native preparations. Vital staining. Whole mounts, sections, squashes, dry mounts. Fixation. Fixatives. Staining. Aqueous media. Non-aqueous mounting media.

- Whole mounts must be prepared to pass the zoological part of the course. Botanical part of the course: active participation, elaboration of three protocols, written test in which 75% of correct answers is needed to pass.
