Course objectives
The main objectives of the course are as follows:
- Introduction to the principles of microscopy: understanding the basic principles of different types of microscopes;
- Mastering different microscopy techniques such as light microscopy, phase contrast microscopy, polarizing microscopy, etc.;
- Preparation of microscopic specimens: students will learn how to prepare specimens appropriately for microscopic observation and how to identify different types of plant cells, tissues and organs;
- Staining microscopic specimens: students will learn the different staining techniques used in staining plant material;
- Creating permanent slides: students will learn the procedures necessary to create permanent microscopic slides;
- Advantages and limitations of different methods: students will understand the advantages and limitations of different microscopy methods and when it is appropriate to use a particular technique;
- Applications in different disciplines: students will learn about applications of different methods in different fields of plant research.
Learning outcomes
At the end of this practical course students should be able to:
describe the development of the light microscope and actual construction of the light microscope;
operate the light microscopes in different modes and techniques;
use various methods for preparations from botanical objects;
document these objects by means of microphotography;
process the microphotographs.

  • Root tip squash Method: Cell Cycle, Mitosis, Chromosome Staining.
  • Anatomical Methods: Fixation. Dehydration. Infiltration and Embedding. Paraffin Sections. Sections Staining and Mounting.
  • Vital Preparations: Phase Contrast Microscopy, Nomarski DIC.

Teaching methods
Presentations covering theoretical bases of different procedures for the study of plant tissue structures using the light microscope are used as an introduction of each lesson. Practical procedures of selected histological techniques lead to microscopic preparation which is analysed by means of proper method of the light microscopy. Plant structure is documented by drawing or as microphotography. Protocol of each procedure containing theoretical base as well as analysed results is prepared by each student as homework.
