- General Biology (programme PřF, N-BI, specialization Microbiology)
- Course objectives
- At the end of the course students should be able to understand principals of work with bacterial cells and bacteriophages and to understand chosen tasks, to solve various problems they can face in bacterial physiology; to use set of chosen techniques used in physiological laboratories; to work with growth characteristics of bacterial cells and phages, influence of temperature and UV on lysogenic cells and phages; to prepare crude enzymatic preparation and to test its enzymatic activity.
- Syllabus
- Determination of the bacterial proteins and dry-weight. Bacterial growth curve in static culture (using plate counts). One-step growth curve of the polyvalent and virulent staphylopage. Determination of dehydrogenase activity. Preparation of the crude enzymatic preparation (HEP) with phosphatase activity. Determination of phosphatase activity of native cells and HEP. Oxygen consumption (Oxi-Top). Biodegradation. Influence of heavy metals on bacterial growth (Bioscreen).
